This has a number of steps, depending on the process structure of your organization.

1. After a build is ready, or on its expected date, the developement leader in charge should officially inform about the readiness (If any exception is needed, it should be addressed and should be confirmed by the respective senior management)

2. From the build folder, usually a shared path or a folder in the configuration management tool like VSS, the testing team lead or the release manager deploy the build so that it is readily available for testing

3. The same person informs the test engineers officially. Here is where the testing process starts. It may start from a sanity testing, to rigorous system testing etc.

NB: As I've said before, the responsibilities varies with roles. For companies up to the medium level, it is often found that a same person needs to assume various roles. Ex: A test lead or a test manager may double as a release manager.
